Feline Leukemia Virus subgroup C Receptor 1 (FLVCR1) is an essential transporter whose mutations are responsible for rare neurodegenerative disorders, including Posterior Column Ataxia and Retinitis Pigmentosa (PCARP), Hereditary Sensory and Autonomic Neuropathy (HSAN), and congenital hydrocephalus (CH). We recently demonstrated that the full-length isoform encoded by the FLVCR1 gene, FLVCR1a, interacts with the IP3R3-VDAC1-GRP75 complex. This complex localizes at the mitochondria-associated membranes (MAMs) that are involved in several processes including calcium flux from the endoplasmic reticulum (ER) to mitochondria. This project will focus on another isoform encoded by the FLVCR1 gene, named FLVCR1b, previously shown to localize in mitochondria. Specifically, the project aims to investigate in further detail the function of FLVCR1b. The results of this project will have important implications for understanding the pathogenesis of FLVCR1-related disorders.
